Induce new hair follicles for baldness – a future personalized hair regeneration method


Columbia University Medical Center and University of Durham scientists collaborated to bring us a step closer to cure baldness (2013, October 21). Bald guys be patient. This step closer to a  cure in mice may reach humans.  Hair regeneration method is first to induce new human hair growth.  Many laboratories are working on curing baldness.  Growing hair follicle cells further in the laboratory are proving frustrating.  This laboratory circumvented that hurdle by using specific cells called dermal papillae cells and figuring out how the cells “speak” to each other and other cells around them to create a hair follicle. For the first time, researchers have been able to take human dermal papilla cells (those inside the base of human hair follicles) and use them to create new hairs. Dermal papillae cells are embedded in a rich, matrix at the base of the hair follicle and are essential for induction and maintenance of hair growth.

The study was published today in the online edition of the Proceedings of the National Academy of Sciences (PNAS) on October 21st, 2013, which you may click here to read. 
The significance of this study, briefly quoted below, is that it has the potential to grow new hair using a patient’s own cells – providing a personalized hair growth solution for baldness, says co-study leader Angela M. Christiano. Additionally, according to co-study leader Dr. Jahoda, this is an important step toward the goal of creating a replacement skin that contains hair follicles for use with, for example, burn patients

Significance

Growth of de novo hair follicles in adult skin occurs by a process known as hair neogenesis. One way of initiating neogenesis is to place dermal papillae isolated from the hair follicle in contact with an overlying epidermis where they reprogram the epidermis to adopt a follicular fate. This approach, however, has not been successful using cultured human dermal papilla cells in human skin because the cells lose their ability to induce hair growth after expansion in vitro. In this paper, we demonstrate that by manipulating cell culture conditions to establish three-dimensional papilla spheroids, we restore dermal papilla inductivity. We also use several systems biology approaches to gain a comprehensive understanding of the molecular mechanisms that underlie this regenerative process.

Similar baldness research in other laboratories
For years, scientists had thought that people suffering from hair loss had a depletion of hair follicles and follicle stem cells, which are necessary to grow hair. Dr. George Cotsarelis, a professor of dermatology at the University of Pennsylvania, published a study showing that bald people have the same number of follicle stem cells as those with hair. So if researchers could identify the signals that stimulate the stem cells into producing more hair follicle progenitor cells, then it would be possible to generate bigger hair follicles that could grow hair. Such studies have shown that men with male pattern baldness still have stem cells in follicle roots but these stem cells lose the ability to initiate hair regeneration.

University of Toronto scientists have discovered cells in hair follicles that seem to function like dermal stem cells and could have potential uses in many areas of medicine.

Whilst nobody currently offer stem cell therapy for hair loss, this research offers insight into how stem cell therapy can be used for a range of skin conditions, including scarring and skin cancer.

Freda Miller, Ph.D., professor of molecular and medical genetics at the University, reviewed her and her colleagues’ current and prior research at a recent International Society for Stem Cell Research meeting.

The research may someday offer stem cell therapy for hair loss.

To make your own filaments for cheaper 3D printing: Invention of Lyman Filament Extruder II


An 83 year old inventor of the Lyman Filament Extruder II, Hugh Lyman, who in 1976 wrote in The Computerworld magazine about IBM 5100, an early 55 lb “portable computer” may have solved a major source of expense for those interested in 3D printing. The 3D printer has been called “The emerging gadget which may very well change everything” by Time Magazine in 2013. A desk-top model of the 3D printer may be available for $500.00.

Answering a call for an invention that would solve the problem of expensive filaments required for 3D printing, Hugh Lyman submitted two entries. More than 12,000 3D printing enthusiasts from around the world have already downloaded the plans of his two extruders. Atleast one 3D company Lulzbot, plans to sell a pre-assembled version. The second entry is a cheaper to make and use version, and won the $40,000 Inventables award, set up by Zach Kaplan, CEO of Inventables. Sperm and Egg Donation: Should the children know their origins before choosing a mate?


Yes, because there are a few donors who are increasingly selected and are quite popular. Which means that a child born from such a donor might very well be meeting a half-sibling often and not know it at all. Now, normally that would not be a major issue. How often do we realize that we do not enjoy the company of our relatives?  We cannot choose our relatives. This situation is truly magnified in a child born from a popular donor sperm. The mothers may very well be from the same town and the children may even be in the same classroom. Yet, the donor himself might have been flown in from a different state for a few hours. This way the donor is not morally or emotionally connected to the results of his donation. This impending tsunami of children born to same sex unions or infertile couples is waiting to happen in twenty years when they grow up and form their own unions. Many of the donor’s childrens’ parents are single men/women, gay or lesbian couples or infertile couples.

“Are you my sibling?” Will that be the first question after “hello”?

Hilarious situations can arise when if in a hypothetical high school classroom a dozen students discover they are half-siblings through a donor. Now imagine the number of happy reunions in which the same sex couples choose to have a donor. Add to this the following scenario: a check list of donors. Why? is that very important? See this chart of rising number of same sex unions. If each union uses a donor and more than one uses the same popular donor do you see what might happen? How do we help a teen be sure that good looking teen is not a sibling from the same anonymous donor? Should we legislate an anonymous number system for those donors who wish to remain anonymous?

How many eggs can a woman donate?
A woman can donate only so many eggs and yet a woman may choose to be a popular donor. Even her most enthusiastic attempts cannot match that of an inspired male sperm donor. Department of Health admits that donors are not well informed. A Columbia university student asks anonymously questions about donating eggs on reading an advertisement on a Columbia Health site, where “Go Ask Alice” addresses her fears and concerns. On this Huffington Post site a woman donor explains how she does not want to be a mother and yet, donating her eggs to a couple who want to have children made her feel less guilty.

How many sperms can a man donate
A man has the potential to donate an unlimited number of sperms. Fortunately, ethical sperm banks will allow the donator to have a maximum of ten children. The sperm donor goes through a stringent selection process as described in this Stanford University list for a sperm donor. Intelligent, virile young men are much in demand as sperm donors to father a child. Not surprisingly, such men are found in universities and so, many such sperm clinics are near universities.

More than a dozen siblings from a sperm donor?
A documentary film “Donor Unknown” documents the search for Donor 150 who is the anonymous donor of more than a dozen children in a small beach community, who discover each other on a web site set up for such descendants. There is an interesting article on Boston.com about this young father who sired more than 70 children through donating his sperm in college. His fiance was a little miffed on finding out about his offsprings on a reality TV show, during which he was introduced to two of his biological children.

If you are at least 5’10”, between the ages of 18 and 35 years, have post-secondary education (college, vocational or technical) and are within normal limits of weight for your muscular build and height, you may be a potential candidate to become a donor. We need donors with all types of racial and ethnic backgrounds. We are not able to accept applicants who use tobacco products in any form, including smoking or ingesting marijuana. Use of these substances will be tested for throughout the donation process.

The quote above is from a real sperm bank that has a several locations. Yet, what if several women selected the same donor at the same location? Well, many children would be born in the same town not knowing they are siblings, unless they had a way to know that.

Donors donate for financial or emotional reasons or both. Legislators need to plan for this impending tsunami of children born to same sex unions or infertile couples who in twenty years will want to be certain they are not dating their sibling, from an anonymous donor.

Worm Theory: to improve the immune system to fight asthma, multiple sclerosis and more


The probiotic way, where you can introduce worms or germs inside the gut by eating them intentionally, is becoming increasingly mainstream (click here). Have we “poisoned” our guts by making it sterile? Have we “killed” our ability to fight invading microbes like common bacterial infections, or usual allergens like pollen in the spring time? Perhaps, we have made our outside environment too clean, say many physicians today. Maybe, you should eat probiotics.

Worms? Yes, eat small worms or tapeworms for a healthier immune system.

When great scientists go unrecognized by the nobel prize: how should the public make up for that lapse?


Any body who has a family member suffering from an auto-immune disease realizes the importance of research in immune biology. Severe allergy, Multiple sclerosis, Rheumatoid Arthritis, Crohn’s Disease, Lyme Disease, Osteoarthritis, and recently, Diabetes are a list of diseases that are related to a malfunction of the immune order in a body. The body begins to attack it’s own cells believing that it is a “foreign” cell or an invading organism that needs to be destroyed. The degeneration of parts of the body begins leading to specific disease. The New York Times covers the story of two scientists from two different countries who collaborated in the same lab to discover answers to the immune system. Read here to learn about: the seminal contributions of immunologists Charles A. Janeway Jr. (1943-2003) and Ruslan Medzhitov. Their work went unrecognized by the nobel committee. Now, you as the public must begin to award and recognize the scientists you value. Shower them with emails and let them know that you care about their research. Let them feel appreciated. Invite them for talks. Give them free dinners at talks – yes, scientists love that free cookie.

The immune system is two fold: The biological clock of the body controls the innate immune system – just like plants, all humans, animals and birds are conditioned by nature to wake up with the sun, have light exposure of several hours and then off to sleep by sunset or dark. Flowers close, birds stop chirping, animals begin to snore, but humans? Night shift workers have higher rates of illnesses. Could it be a biological clock gone awry?
The innate immune system controls the adaptive immune system which controls how we respond to microbes and environmental insults. When the biological clock is off, the innate immune system is off and since it controls the adaptive immune system, the response to body insult is off. Any wonder why unhappy people or those who work irregular hours feel sicker?

Which scientists should the public honor and thank for researching the innate and adaptive immune system and the way they are connected? You must read the following
A revolutionary idea of how the immune system worked. In vertebrates, two immune systems protect against infection. There’s the evolutionarily older system that all organisms, even plants, have — the innate immune system. There is also another system that evolved relatively recently, the adaptive immune system, which only vertebrates have. Janeway proposed that the innate system detects invading microbes, distinguishes them from everything else and signals the adaptive immune system to move against the intruder.

Is it time to revamp intellectual property world rights and how they are implemented?


The internet has a way of democratizing knowledge.  It has allowed free access to knowledge anywhere in the world by anybody who has a computer. When Napstar, an upstart music sharing software program allowed unprecedented music download and sharing, it forever changed the way copyrights were viewed and protected. Initially, intellectual property rights were created by industries to protect their rights to research, develop and sell with a monopoly for a certain number of years a product developed with the intellectual property rights giving them protection for a few years to recoup investment.

Now, the little guy can access that knowledge and dispense it freely. What is the direction of international intellectual property protection policy to be adopted? Should policy work towards an environment encouraging more innovation which comes only from uninhibited sharing of world – wide knowledge freely? Or should it restrict sharing of content at prohibitive costs, and discouraging free sharing of knowledge? Should policy protect the investments towards research and development which has greatly increased the life span of mankind this century, and allowed automobiles to connect mankind to ends of the earth? While this has ensured large profits for the industrialized, developed world, it has benefited many. Or has it? What about the developing country’s Shamans that freely shared their ancient medicinal knowledge with the industrial developers to isolate the active chemical compound from? Did the large company ever “take care” or continue to compensate for the life of the patent the barefoot shaman in an an impoverished country who with a toothless smile shared his timeless knowledge, perhaps for free or perhaps even offering their hospitality?

Such questions were asked in the past but the answers shrugged away. However, this recent 2009 -2012 world-wide recession has brought the world together through the internet with forums discussing international needs and internation humanitarian conditions and world-wide travel and space travel and border rights and free music and hollistic medicine and mixed marriages and mixed gene pools and air rights and water rights and more. Everywhere there are youngsters understanding the international language of computers in a way their educated, elders could not imagine even today.

What is this world of the future? Who are to make the new rules? How do we continue to serve the interests of innovation and progress with world-wide dissemination of knowledge best? Should knowledge be free?

In fact, it is remarkable to consider that the areas where the Internet has succeeded most readily – for example as a giant distributed database of facts on any subject under the sun – are traditionally those in which there are little or no intellectual property rights. The software on which the Internet runs is largely open source, another Internet-enabled method of innovation to which policy makers have been slow to adapt. The Internet offers us remarkable opportunities to achieve the real goals that intellectual property policy ought to serve: encouraging innovation and facilitating the dissemination of cultural and educational materials. Yet policy making has focused almost entirely on the Internet’s potential for illicit copying. An example demonstrates the point.
• Copyright term limits are now absurdly long. The most recent retrospective extensions, to a term which already offered 99% of the value of a perpetual copyright, had the practical effect of helping a tiny number of works that are still in print, or in circulation. Estimates are between 1% and 4%. Yet in order to confer this monopoly benefit on a handful of works, works that the public had already “paid for” with a copyright term that must have been acceptable to the original author and publisher, they deny the public access to the remaining 96% of copyrighted works that otherwise would be passing into the public domain. Before the Internet, this loss – though real – would for most works have been largely a theoretical one. The cost of reprinting an out-of-print book or copying and screening a public domain film was often prohibitive. But once one adds the Internet to the equation, it becomes possible to imagine digitizing substantial parts of the national heritage as it emerges into the public domain, and making it available to the world. Now this is truly fulfilling the goals of copyright: encouraging creativity, and encouraging access.

Intellectual property firms are collapsing in 2012. The old model is no longer apparently working with a botique IP firm serving the needs of many. Preclinical studies: Autism compound, KM-391, significantly decreased plasticity of the brain and increased serotonin levels


Cellceutix is developing an experimental drug, KM – 391, to treat autism.  Cellceutix, is a biopharmaceutical company developing small molecule drugs to treat diseases like Autism, Psoriasis and Cancer. KM-391Clinical Trial Data in Animals

Preclinical studies have shown that administration of the autism compound, KM-391, significantly decreased plasticity of the brain and increased serotonin levels in all three regions of the brain: cerebral cortex, hippocampus, and caudate nucleus, from very low levels, as seen with 5, 7-Dihydroxytryptamine (5, 7-DHT)-induced autism, to normal levels, as observed in placebo-treated control without the neurotoxin 5, 7-DHT.

Cellceutix recently acquired the small molecule technology and began preclinical trials in animals. They conducted three studies, which you may click here to read in detail on Cellceutix site. It includes:

3. KM-391 Third Study – Serotonin Levels Measured in Three Different Regions of the Brain.

“These results support KM-391 as a possible therapeutic agent in the treatment of autism in patients,” Mr. Ehrlich contends. “If we can deal with the core issues of autism and not just the symptoms of the disease, it would be a major breakthrough and offer hope to families and patients who suffer from autism.”

Based on results from a third animal study showed KM-391 was a possible therapeutic agent in the treatment of autism in patients, CEO Leo Ehrlich says Cellceutix is working on possible collaborations to further develop the autism compound, which can be quite expensive.  It has a memorandum of understanding with a company in India, able to financially support and implement all the pharmacology, preclinical work, including cGMP manufacturing, and toxicology, in compliance with U.S. IND requirements.

On November, 2011 announced Cellceutix Chief Scientific Officer Dr. Krishna Menon, “We are extremely pleased to add Dr. Marks to our team. We have assembled a world-class advisory board with Dr. Emil Frei, Dr. Samuel Danishefsky, Dr. Paul Ginsburg and now Dr. Paul Marks. Going forward with Kevetrin™ in clinical trials, as well as other drugs in our pipeline, their experience and knowledge will play a key role in our success”. Dr. Marks served as President and Chief Executive Officer of Memorial Sloan-Kettering Cancer Center (“MSKCC”) from 1980 to 1999. His addition to the board shows the firms commitment is towards their cancer drug, also currently in the pipeline.

High Fructose corn syrup – why would anyone educated still consume it?


Two scientifically researched and published papers show the connection between consumption of high fructose corn syrup and NAFLD. One is a review published in Nature reviews and the other a research paper of a three year study. A third research group says that high fructose corn syrup should be declared an environmental toxin. A fourth paper publised by a team of nutritionists in the American Journal of Clinical Nutrition in 2004, suggest that the obesity epidemic trend since 1970s is closely related to 1000 fold increase in use of high fructose corn syrup in sweetened beverages. 1) The review postulates that the Western diet which includes excessive fructose may be leading to Nonalcoholic fatty liver disease (NAFLD). Which means that liver damage from excessive consumption of alcohol is expected. However, nonalcoholics are often diagnosed with liver damage. These reviewersblame their western diet which includes beverages sweetened with high fructose corn syrup. The damage is in two steps and the second step can be prevented with antioxidants. However, the western diet does not include or is deficient in antioxidants. The combination of high fructose and lack of antioxidants may be contributing to the most frequent liver disease worldwide – NAFLD. 2) Another group of researchers (Liver Unit, Ziv Medical Center, Safed, Israel and other colleagues)were perplexed when faced with patients with fatty liver when they did not have the traditional risk factors such as obesity, diabetes and hyperlipidemia. So, they conducted a three year, systematic scientific research study with patients with fatty liver and healthy people (controls) with no fatty liver. They used a validated food questionnaire given by a dietician. They followed the daily intake of food and beverages and sources of added sugar. Their results?

80% of the patients with fatty liver consumed sweetened beverages compared to 20% of healthy people. They included 50grams per day of added sugar to their beverages which was considered excessive by the researchers and 40% of the patients with fatty liver consumed four of more of such beverages which were excessively sweetened. Ultrasound findings revealed mild fatty liver in 44% of the cases to severe fatty liver in 18% of the cases.

Soft drink beverage was the only independent variable able to predict a fatty liver in 82.5% of cases.

Their conclusion is obvious isn’t it? Change your beverage habit. Unsweeten it. If you cannot stop it yourself, just do not introduce your child to sugar or corn syrup sweetened beverages from childhood. The child will never crave a sweetened beverage and prefer plain water, like our healthier ancestors.

3) A third group of researchers say that high fructose corn syrup depletes ATP and causes uric acid to collect in human bodies. They say it is an environmental toxin. Write to this research team to learn more or if you wish to encourage them for further research on the effect of fructose on renal biology and disease.

The child will probably never crave a sweetened beverage and prefer plain water, like our healthier ancestors. You may want to write to the pediatricians and scientists listed above and ask why they are concerned of this rising public health threat. The more informed you are, the better you can serve your community and prevent it from facing cripling medical expenses in the near future.

Why and how to add Calcium in the diet of a person with Autism Spectrum Disorder


Cells in the brain contain proteins called calcium channels that can regulate Calcium levels within the cells. Calcium levels inside the cells are 10,000 fold lower than Calcium levels outside the cells. The calcium channels are gate keepers of Calcium, allowing only the required amount inside the cell by opening briefly and then the gate shuts. Calcium cannot be changed or transformed by the body, which makes it imperative to regulate calcium levels inside the cells, to prevent it’s build up at toxic levels.

Gene mutation: A mutation in the gene for these gate proteins can alter their structure causing them [calcium gates] to stay open much longer than usual. This allows calcium to continue to enter inside the cell abnormally. The result? An overload of Calcium inside the cell. It is a novel disorder characterized by autism and other features. Inheritance of calcium channel disorder: Most cases of this calcium channel (CaV1.2) disorder is inherited by a new mutation (G406R) with no previous history of this altered gene in their family.

Possible therapy? Although, the role of this calcium channel as important for the contraction of the heart is known, its role within the cell is not well understood, yet. The disorder is caused by an inability of these Calcium gates to close. What does that result in? Yes, a Calcium overload inside the cells. This also suggested to the researchers that Calcium channel blockers may be useful for treating this disorder.

Researchers: This discovery by Dr Igor Splawski and colleagues demonstrates the importance of Calcium imbalance and established the importance of Calcium in human development and implicates Calcium in cell signaling and Autism.

Although, the gene mutation causes this disorder, it is also clear that too much Calcium cannot be good for a human. That is why, it should not surprise anyone who has read Dr Igor Splawski’s teams’ research papers to learn about other issues when a person eats too much Calcium. You will also learn that they regularly have high doses of Calcium supplements to prevent osteoporosis or bone loss in women as they age. It is unclear why eating natural sources of Calcium is safe while eating Calcium in excessive supplement form might cause bones to break.
